How it Works: | ||||||||||||||||||||||||||||||||||||
Conjugate Diluents stabilize newly conjugated proteins, reconstitute yophilized conjugates, and dilute concentrated conjugates. Cell Technologys conjugate diluents stabilize the immunoglobulin and HRP components of the conjugate - protein complex. All conjugate diluents are available at 1x and 5x concentrations. When stored unopened and protected from light, a shelf-life of 18 months can be expected at 2°-8°C.

Directions: | ||||||||||||||||||||||||||||||||||||
To stabilize newly conjugate proteins, add 1 part of the appropriate 5x conjugate diluent (that has not been diluted to 1x) to 4 parts of the new conjugate solution.
To reconstitute lyophilized conjugates, simply pipette a known amount of a 1x conjugate diluent into the vial. The liquid conjugate is now ready to be used directly, or further diluted to the required concentration. To dilute concentrated conjugates for use in an assay, pipette a known amount of the conjugate stock into a 1x conjugate diluent. |

ECD100 Monoclonal- Polyclonal Conjugate Diluent & Stabilizer
Monoclonal-Polyclonal Conjugate Diluent & Stabilizer is a unique formulation specifically designed for use in mouse monoclonal capture and polyclonal-HRP sandwich ELISA formats. This diluent contains mouse proteins which suppress nonspecific binding of the polyclonal-HRP conjugate to the mouse immunoglobulins adsorbed onto the plate. These protein additives also provide additional structural stability to the conjugate protein complex. Other additives contained in ECD100 serve to extend the useful titer of the conjugate by stabilizing both the immunoglobulin and HRP components of the conjugate. Antimicrobial agents have also been added to retard bacterial growth. When stored unopened and protected from light, a shelf-life of 18 months can be expected at 2°-8°C.

ECD200 Antigen-Down Conjugate Diluent & Stabilizer
Antigen-Down Conjugate Diluent & Stabilizer is a unique formulation specifically designed for use with anti-IgG-HRP conjugates. This diluent contains proprietary non-mammalian protein additives which will not interfere with anti-IgG-HRP conjugates used in antigen-down and sandwich ELISA formats. These additives also provide additional structural stability to the IgG-HRP conjugate protein complex. CD2 will also inhibit conjugate binding to nonspecific serum proteins bound to the plate, thus reducing conjugate bridging on the plate surface and lowering background noise. Antimicrobial agents have been added to retard bacterial growth. When stored unopened and protected from light, a shelf-life of 18 months can be expected at 2°-8°C.

ECD300 Polyclonal-Polyclonal Conjugate Diluent & Stabilizer
Polyclonal-Polyclonal Conjugate Diluent & Stabilizer is a unique formulation specifically designed for use in polyclonal capture and polyclonal-HRP sandwich ELISA formats. Proprietary additives were specifically selected to provide optimal assay performance when using rabbit, goat, or rabbit and goat polyclonal antibodies in a sandwich ELISA format. ECD300 contains a uniquely processed form of rabbit and goat IgG to suppress nonspecific binding of the polyclonal-HRP conjugates to the plate surface. To reduce nonspecific binding interactions, a detergent is also included. Antimicrobia agents have been added to retard bacterial growth. When stored unopened and protected from light, a shelf-life of 18 months can be expected at 2°-8°C.

ECD400 Monoclonal - Goat Polyclonal Conjugate Diluent & Stabilizer
This diluent is a unique formulation specifically designed for use in monoclonal capture and goat polyclonal-HRP sandwich ELISA formats. Additives were carefully selected to specifically address the stability and nonspecific binding issues associated with this type of ELISA format. Goat proteins are included to reduce any nonspecific interactions between the monoclonal capture and goat polyclonal-HRP antibodies. CD4 also contains mouse IgG to specifically suppress nonspecific bridging of the goat polyclonal-HRP conjugate with the mouse capture IgG on the plate surface. To further reduce nonspecific binding of the conjugate to the plate surface, adetergent is also included. Antimicrobial agents have been added to retard bacterial growth. When stored unopened and protected from light, a shelf-life of 18 months can be expected at 2°-8°C.

ECD600 Conjugate Stock Stabilizing Reagent
Conjugate Stock Stabilizing Reagent contains a BSA-based mammalian protein additive to stabilize conjugated proteins, and magnesium and calcium salts to stabilize the catalytic site of the HRP porphyrin ring structure. When diluted to 1x, ECD600 is an excellent HRP conjugate diluent and stabilizer base, to which additional components can be added to for use in your specific assay.
At 5x, ECD600 is an excellent diluent for stabilizing newly conjugated proteins. This allows the liquid conjugate stock to be stored, and then later diluted to make a working conjugate for use in the assay. Simply add 1 part of 5x CD6 (that has not been diluted to 1x) to 4 parts of the new conjugate solution. For example, if you just eluted 20 mL of an antibody-HRP conjugate off a column, simply add 5 mL of 5x CD6 to the conjugate solution. If the conjugate was initially at 10 mg/mL, the stabilized conjugate will now be at 8 mg/mL in 25mL diluent (its been diluted by 20%). Antimicrobial agents have also been added to ECD6 retard bacterial growth. When stored unopened and protected from light, a shelf-life of 18 months can be expected at 2°-8°C. |
